چکیده

Breast cancer is one of the most common malignancies in the world. According to the global cancer statistics, Europe and America have the high incidence and mortality (Parkin et al., 2005). The incidence of breast cancer is 20 per 100,000 populations, and the incidence is growing (Zheng et al., 2001). Research has shown that about 20% 30% of the breast cancer patients have HER-2/neu amplification or overexpression, which is associatedf with a more aggressive phenotype and decreased survival (Paterson et al., 1991; Seshadri et al., 1993; Press et al., 1993; 1997; Ross and Fletcher, 1999). The benefit of humanized anti-HER-2/neu monoclonal antibody trastuzumab (Herceptin) in HER-2/ neu -positive breast cancers has been well documented as noted by prolonged survival (Dean-Colomb and Esteva, 2008). But this therapy is effective only if the detection of HER-2/neu status is accurate. There are several methods available to detect the HER-2/neu status like polymerase chain reaction (PCR), immunohistochemistry (IHC), fluorescence-in-situ hybridization (FISH), Chromogenic in situ hybridization (CISH) (Tanner et al., 2000; Sáez et al., 2006). Protein overexpression detected by IHC or amplification of HER-2/neu gene analyzed by FISH are the two main methods used to detect HER-2/neu status

Fluorescent in Situ Hybridization and Real-Time Quantitative Polymerase Chain Reaction to Evaluate HER-2/neu Status in Breast Cancer

Background:Breast cancer remains the most common and second lethal cancer in females. HER-2/neu is one of the most important amplified oncogene in breast cancer. The amplification of HER-2 is correlated with decreased survival, metastasis, and early recurrence.  The amplification of HER-2/neu gene and synthesis of th...

FISH for HER-2/neu in breast cancer: standardization makes the difference!

CONTEXT Overexpression of HER-2/neu oncogene in breast cancer patients is correlated with disease free survival (DFS) and overall survival (OS). The most commonly used methods for the detection of HER-2/neu status are immunohistochemistry (IHC) and fluorescence in situ hybridization (FISH). However, therse is a lot of controversy with regard to the best method. Most of the FISH studies chose ar...

Evaluation of HER-2/neu status in breast cancer specimens using immunohistochemistry (IHC) & fluorescence in-situ hybridization (FISH) assay

BACKGROUND & OBJECTIVES Fluorescence in situ hybridization (FISH) is increasingly being recognized as the most accurate and predictive test for HER 2/neu gene amplification and response to therapy in breast cancer. In the present study we investigated HER-2/neu gene amplification by FISH in breast carcinoma tissue specimens and compared the results with that of immunohistochemical (IHC) analysi...

Laboratory assessment of the status of Her-2/neu protein and oncogene in breast cancer specimens: comparison of immunohistochemistry assay with fluorescence in situ hybridisation assays.

AIM To evaluate the clinical usefulness of three commercially available assays for Her-2/neu oncogene and protein measurements. The Her-2/neu protein is overexpressed, mostly as a result of gene amplification, in 20-30% of human breast cancers, and has been shown to have prognostic and predictive value for treatment with chemotherapy or the new monoclonal antibody, Herceptin. METHODS An immun...

Assessment of HER-2/neu status in breast cancer using fluorescence in situ hybridization & immunohistochemistry: Experience of a tertiary cancer referral centre in India.

BACKGROUND & OBJECTIVES Determination of HER2 status in breast cancer has become important to identify potential candidates for anti-HER2 therapy. In this study we compared fluorescence in situ hybridization (FISH) and immunohistochemistry (IHC) for the determination of HER2 status in breast cancer patients referred to a tertiary care referral centre. METHODS A total of 200 cases of invasive ...
